Biochem/physiol Actions: Two-chain polypeptide hormone produced by the β-cells of pancreatic islets. Its molecular weight is ~5800 Da. The α and β chains are joined by two interchain disulfide bonds. The α chain contains an intrachain disulfide bond. Insulin regulates the cellular uptake, utilization, and storage of glucose, amino acids, and fatty acids and inhibits the breakdown of glycogen, protein, and fat.
Reconstitution: Insulin has low solubility at neutral pH. It can be solubilized at 2 mg/ml in dilute acetic or hydrochloric acid, pH 2-3.

Application:
Insulin is used as a growth factor in many mammalian cell culture systems.
• Insulin from bovine pancreas has been used for the differentiation of MSCs (mesenchymal stem cells) to adipocytes.
• It has been used for culturing of various cells.
• It has also been used to study insulin-mediated inhibition of the HMGCS2 (3-hydroxy-3-methylglutaryl-CoA (HMG-CoA) synthase) gene.
